Structural insights into sGC activation by different activators

چکیده
Background The soluble guanylyl cyclase (sGC) is a key enzyme involved in the production of the second messenger cGMP. Due its cardiovascular relevance, sGC has been the target of numerous drug discovery efforts leading to the development of many different activators and stimulators with pharmaceutical potential. One of such compounds is the sGC activator Cinaciguat (BAY 58-2667) which is in clinical trials for acute decompensated heart failure.

New insights into the role of soluble guanylate cyclase in blood pressure regulation.
PURPOSE OF REVIEW Nitric oxide (NO)-soluble guanylate cyclase (sGC)-dependent signaling mechanisms have a profound effect on the regulation of blood pressure (BP). In this review, we will discuss recent findings in the field that support the importance of sGC in the development of hypertension. RECENT FINDINGS The importance of sGC in BP regulation was highlighted by studies using genetically...
متن کاملStructural insights into sGC
The membrane and soluble guanylyl cyclases (sGCs) are key families of enzymes that produce the second messenger cGMP. Although both families recognize different ligands and have therefore different N-terminal input domains, they do have similar C-terminal output domains containing a coiled-coil domain and a guanylyl cyclase catalytic domain. We present here our latest structure-function studies...
متن کاملCharacterization of two different five-coordinate soluble guanylate cyclase ferrous-nitrosyl complexes.
Soluble guanylate cyclase (sGC), a hemoprotein, is the primary nitric oxide (NO) receptor in higher eukaryotes. The binding of NO to sGC leads to the formation of a five-coordinate ferrous-nitrosyl complex and a several hundred-fold increase in cGMP synthesis. NO activation of sGC is influenced by GTP and the allosteric activators YC-1 and BAY 41-2272. Electron paramagnetic resonance (EPR) spec...
متن کاملPrimary response of the sGC heme binding domain to the cleavage of the Fe-His bond
Soluble guanylate cyclase (sGC) is an important heme sensor protein. Regulation of the status of heme in the heme binding domain (or HNOX domain) by various gaseous activators can increase the catalytic efficiency of the cyclase domain. Several studies have demonstrated that the full activation of sGC is directly related to the cleavage of the Fe-His bond of the HNOX domain. To expand the prima...
متن کاملMechanism of YC-1-induced activation of soluble guanylyl cyclase.
The signaling molecule nitric oxide (NO) mediates many of its effects by the stimulation of soluble guanylyl cyclase (sGC). The activation process is initiated by high-affinity binding of NO to the enzyme's prosthetic heme group. Despite its poor sGC-activating properties, carbon monoxide (CO) has also been suggested as a physiological activator of sGC. Recently, we have shown that the substanc...
